Studies in heart and brain pointed to an important role for plasmalemmal and, later, mitochondrial ATP-sensitive potassium channels (KATP) in the acquisition of early tolerance. It has been demonstrated that rapid induction of tolerance in brain involves a cascade of events triggered by the liberation of adenosine in response to ischemia, stimulation of adenosine A1 receptors, and opening of plasmalemmal KATP , which leads to membrane hyperpolarization and reduced neuronal excitability (41). Further supporting this finding, ischemic tolerance in hippocampal neurons in gerbils was blocked by adenosine-receptor antagonists (42).
