Lubeluzole

The exact mechanism of action of lubeluzole, a drug effective in animal models, is unclear. It might block sodium channels in cells, and it might reduce the release of nitric oxide, a neu-rotransmitter generated by activation of the NMDA receptor. Post hoc analysis suggested that the drug was beneficial in certain age and stroke severity groups in initial Phase 2 clinical trials. However, a Phase 3 confirmatory trial did not show that lubeluzole was efficacious in this subset of acute stroke patients, and further stroke research with this drug has been abandoned (14), illustrating the danger in placing too much emphasis on subgroups analyses.
